abstract | [PROBLEMS] To provide a novel screening system capable of searching for compounds involved in the Ras / MAPK pathway or Ca 2+ / CN pathway, or compounds that modify human Th1 / Th2 differentiation induction under conditions in which those pathways move. . SOLUTION: Human naive CD4T cells are cultured under stimulation with an anti-CD3 antibody in a differentiation induction medium (Th1 differentiation induction conditions) containing IL-2, IL-12 and anti-IL-4 antibodies, or IL- 2. Culture under stimulation with anti-CD3 antibody in differentiation induction medium (Th2 differentiation induction condition) containing IL-4 and anti-IFNγ antibody, and further, anti-IL-4 antibody or anti-IFNγ antibody from the differentiation induction medium, respectively. A method for inducing Th1 and / or Th2 differentiation of human naïve CD4 T cells, further comprising culturing without stimulation with an anti-CD3 antibody under culture conditions excluding only the above.
