abstract When a wild-type actinomycete H + -PPase gene is introduced into Escherichia coli in the form of a plasmid, the transformed strain grows very badly and the plasmid cannot be stably present. In addition, homologous recombination often occurred, resulting in loss of H + -PPase function. A rare codon for Escherichia coli is replaced with another codon on the condition that the amino acid sequence of the wild-type actinomycete H + -PPase gene sequence does not change. The improved actinomycetes H + -PPase gene thus modified can be stably present on a plasmid in E. coli, and the growth of the transformant is not different from usual. The activity of actinomycetes H + -PPase expressed in this Escherichia coli is maintained. [Selection] Figure 5
