abstract | The present invention provides a novel nucleic acid amplification method with increased specificity for a target nucleic acid sequence. A nucleic acid amplification method includes a first nucleic acid complementary to a first sequence with respect to a target nucleic acid having a first sequence, a second sequence, and a third sequence between the first sequence and the second sequence. And a second step of synthesizing a first amplified double-stranded nucleic acid having a third sequence and a predetermined sequence, a second step, and a second step complementary to the second sequence. A third step of exonuclease treatment of the product of the above, a third step of separating the first amplified double-stranded nucleic acid into a single-stranded nucleic acid with a predetermined sequence, and a single-stranded nucleic acid with a predetermined sequence, A fourth step of binding a third primer complementary to the predetermined sequence, a fourth primer complementary to the third sequence, and a fifth step of synthesizing the second amplified double-stranded nucleic acid.
