abstract To provide a TCR β chain specifically recognizing MART-1 27-35 (AAGIGILTV), which is a melanoma specific peptide, and its gene. Peripheral blood mononuclear cells are collected from a patient, and dendritic cells pulsed with a MART-1 A2 peptide are added at a ratio of 1:10 to 1:40 in the presence of a low concentration of IL-2 twice. I was stimulated. In order to further increase the proportion of cytotoxic T cells, T2 cells pulsed with MART-1 A2 peptide were further added to the cell line at a ratio of 1:10 for stimulation. Cloning of MART-1 A2 tetramer-specific cytotoxic T cells is performed by magnetically activated cell sorting of cytotoxic T cells using monoclonal antibodies based on TCR β chain specific antibodies, and DNA sequencing of TCR β chains.
