abstract | The present invention provides a novel enzyme-labeled tyramide that can be effectively used in a signal amplification method (TSA method) using tyramide. A signal amplification method using the enzyme-labeled tyramide is also provided. Tyramide is labeled with thermostable alkaline phosphatase or thermostable peroxidase to form thermostable enzyme-labeled tyramide. One of the signal amplification methods of the present invention can be carried out using the thermostable enzyme-labeled tyramide by the following steps; (1) a primary labeled molecule (for example, biotin) that specifically binds a target molecule to the molecule. (2) a step of binding a peroxidase-labeled primary conjugate to a primary label molecule (for example, a biotin-labeled molecule) bound to the target molecule, and (3) heat resistance in the presence of hydrogen peroxide. A step of reacting a sexenzyme-labeled tyramide or a functional equivalent thereof, and (4) a step of reacting the thermostable enzyme with a chromogenic substrate. [Selection figure] None |
