abstract | 【Task】 Provided is a novel raw starch degrading enzyme produced by an earthworm. [Solution] After suspending the lyophilized powder of earthworm in a buffer and centrifuging, the supernatant is recovered, and after adding a protease inhibitor to the supernatant and ultracentrifugation, ammonium sulfate is added to the supernatant to 35% saturation. The resulting precipitate is dissolved in a small amount of buffer, subjected to anion exchange chromatography using the degradation activity for starch or raw starch as an indicator, and has two raw starch-degrading enzyme activities (elution first) The active fraction was designated as raw starch degrading enzyme I, and the active fraction eluting later was designated as raw starch degrading enzyme II). These fractions were subjected to gel filtration chromatography and hydrophobic chromatography, respectively, and the active fractions were collected to obtain uniform raw starch-degrading enzyme I and raw starch-degrading enzyme II produced by the earthworm.
