abstract Disclosed is a method for producing a polypeptide that can suppress degradation of a target protein and that does not affect the cleavage efficiency depending on the amino acid sequence immediately after the cleavage site, and that uses a method for decomposing a novel tag and the target protein. Provided is a method capable of producing a polypeptide having the same amino acid and three-dimensional structure as a protein. A fusion polypeptide comprising a tag amino acid sequence, a GLD protease cleavage amino acid sequence and a target amino acid sequence is prepared, GLD protease is allowed to act on the fusion polypeptide, and the target amino acid is cleaved by the GLD protease cleavage amino acid sequence. A method for producing a polypeptide, comprising recovering a polypeptide containing a sequence. A method for producing a polypeptide, comprising culturing a host transformed with a DNA encoding Congelin II and an expression vector containing a DNA encoding a target amino acid sequence downstream of the DNA, and recovering the polypeptide having the target amino acid sequence.
