abstract | The present invention relates to oligonucleotides comprising molecular switches that can be in an “open” position or a “closed” position. The molecular switch portion of the probe is particularly sensitive to the identification of sequences complementary to the molecular switch. Oligonucleotides containing molecular switches are applicable to all kinds of hybridization processes. Due to the sensitivity of this oligonucleotide switch domain, probes containing molecular switches are particularly useful for identifying single point mismatches. More particularly, one part (but not all) of the oligonucleotide becomes unbound from the mismatched target. The invention further relates to methods for using these oligonucleotides for research reagents and clinical diagnostics. An exemplary oligonucleotide includes a first hybridization domain, a second cross-linking block domain, and a third binding domain. |
