abstract | The present invention is directed to an improved process for producing modafinil. In that process, benzhydrylthioacetamide is produced in high yield and purity by reaction of haloacetamide with the reaction product of thiourea and benzhydrol in an aqueous solution. This reaction with haloacetamide is carried out in a solvent comprising a basic salt such as potassium carbonate dissolved in water and an organic solvent such as dimethylformamide. The resulting benzhydryl thioacetamide can be oxidized to obtain medafinil for pharmaceutical use. |
