titleOfInvention | Labeling substance for mass spectrometry and phosphorylation position analysis of phosphorylated protein |
abstract | The present invention relates to 1) a step of dephosphorylating one or more serine residues and / or threonine residues of a phosphorylated protein or phosphorylated peptide; 2) the dephosphorylated amino acid residue of the above step 1 is RLG. Wherein R is a nucleophilic functional group that selectively binds to a dephosphorylated amino acid site, and G is a parent-positive child that induces cation formation. The functional group, L is a linker that links the nucleophilic functional group and the parent positive child functional group, step; and 3) the protein or peptide derivative labeled in step 2 is detected by cation mass spectrometry. A method for mass spectrometry or phosphorylation position analysis of a phosphorylated protein or phosphorylated peptide comprising a step, wherein a guanidino group is used as a parent positive child acting group (G) for the labeling substance in Step 2. That it is related to labeling reaction labeled substance of mass spectrometry methods or phosphorylated location analyzing method of phosphorylated protein or a phosphorylated peptide and the analytical method. By using the labeling substance of the present invention, the analytical sensitivity is superior to existing analytical methods, and the phosphorylation position inside the peptide can be analyzed accurately. It can be usefully used for research on functions in the body and diagnosis of numerous diseases. |
