abstract The present invention is a general method for improving the performance of DNA-based vaccines. This method utilizes a complex of DNA and a generalized profile of the antigen to enhance the effectiveness of the DNA-based vaccine and broaden the immune response. A broad immune response improves the protection of the receptor from divergent (but related) strains of pathogens. Furthermore, it effectively improves the effectiveness of DNA-based vaccines used in the treatment of viral diseases including acquired immune deficiency syndrome (AIDS). In one embodiment where the target viral pathogen is HIV (a causative agent of AIDS), this method identifies an ordered group of plasmids with related sequences and uses that plasmid to virus antigens with restricted HLA. A broad and powerful immune response is primed. Therefore, this plasmid mixture can prime an appropriate immune response to reduce the viral burden of HIV-infected persons or to protect uninfected patients from HIV infection.
