The methods, devices and compositions disclosed herein relate to the detection, identification and / or sequencing of biomolecules such as nucleic acids or proteins. In certain embodiments of the invention, an encoded probe comprising a probe molecule attached to one or more nano barcodes can be bound to one or more target molecules. After binding to and separating from unbound coded probes, the bound coded probes can be aligned to the surface and analyzed by scanning probe microscopy. The nano barcode may be any molecule or complex that can be distinguished by SPM, such as carbon nanotubes, fullerenes, submicrometer metal barcodes, nanoparticles, or quantum dots. If the probe is an oligonucleotide, adjacent encoded probes hybridized to the target nucleic acid may be ligated together prior to alignment and SPM analysis. Also disclosed herein are compositions comprising an encoded probe. A system for biomolecule analysis may include an SPM instrument and at least one encoded probe attached to a surface. |
