abstract | PROBLEM TO BE SOLVED: To provide a method for obtaining a gene amplification host cell having versatility, simply and rapidly, and a mammalian cell obtained by such a method. The present invention provides a mammalian cell having a plurality of sites into which a vector containing any foreign gene can be inserted by specific site-specific recombination. Specifically, according to the present invention, an expression vector containing a first site-specific recombination sequence and a selection marker is introduced into a mammalian cell, and the gene and nucleic acid sequence contained in the vector correspond to the selection marker. Gene amplification is performed using a gene amplification inducer, and then a vector containing a second site-specific recombination sequence and a foreign gene is introduced, and the first site-specific recombination sequence and the second site-specific combination are introduced. Mammalian cells produced by site-specific recombination with a replacement sequence by the action of a site-specific recombination inducer are provided. [Selection figure] None |
