abstract PROBLEM TO BE SOLVED: To provide a nucleic acid amplification method with improved specificity for a novel target nucleic acid sequence. A nucleic acid amplification method includes a first nucleic acid complementary to a first sequence with respect to a target nucleic acid having a first sequence, a second sequence, and a third sequence between the first sequence and the second sequence. And a second step of synthesizing a first amplified double-stranded nucleic acid having a third sequence and a predetermined sequence, and a first amplification. A third step of separating the double-stranded nucleic acid into a single-stranded nucleic acid with a predetermined sequence; a third primer complementary to the predetermined sequence with respect to the single-stranded nucleic acid with a predetermined sequence; and a complementary to the third sequence A fourth step of binding to the fourth primer, and a fifth step of synthesizing the second amplified double-stranded nucleic acid. Furthermore, the fourth step is performed by adding a competing nucleic acid that competitively binds to the first primer with respect to the first sequence. [Selection] Figure 3
