abstract | 【Task】 A nucleic acid construct is produced by linking a plurality of nucleic acid molecules without using a restriction enzyme. [Solution] The present invention includes the following: (a) a step of amplifying a first nucleic acid molecule to obtain a first double-stranded amplification product; (b) a second nucleic acid molecule being amplified to obtain a second double-stranded amplification product Wherein the 3 ′ end portion of the first double-stranded amplification product and the 5 ′ end portion of the second double-stranded amplification product are hybridized only with a specific sequence. (C) mixing the first and second double-stranded amplification products obtained by steps (a) and (b), and A step of denatured into a strand and hybridizing under a condition in which only the specific sequence hybridizes; and (d) a nucleic acid amplification reaction using the mixture of (c) as a template to obtain a linked nucleic acid amplification product.
