PROBLEM TO BE SOLVED: To provide a gene replacement vector for replacing avian genes and a method of using the gene replacement vector in order to realize a system for producing foreign proteins in the yolk of birds such as chickens. It is intended to provide. The gene replacement vector of the present invention includes a gene encoding a part of a chicken-derived vitellogenin protein containing a cathepsin D recognition region, and a gene encoding an exogenous EGFP protein is contained in the cathepsin D recognition region. The gene is inserted so as to have a base sequence encoding a cathepsin D recognition region at the 3 ′ end and 5 ′ end of the gene. When the EGFP gene was introduced into chicken liver cells (LMH) using the gene replacement vector, an EGFP protein having a cathepsin D recognition region at the N and C terminus was produced as a vitellogenin fusion protein.
