PROBLEM TO BE SOLVED: To provide a method for rapid identification and characterization of a binding partner for a target molecule, and for providing a binding partner with improved binding affinity. [Solution] Process, (I) contacting a target molecule (TM) having a target first site and a second site with a plurality of first small organic ligand candidates, wherein the TM is a target first site or Including or modified to include a reactive nucleophile or electrophile near the first site, the candidate having a functional group reactive with the nucleophile or electrophile; (Ii) identifying a first ligand derived from the complex of (i); and (iii) designing a derivative of the first ligand identified in (ii) to produce a nucleophile or nucleophile on the TM.
