abstract | Since the method is complicated and there are differences among testers, it is difficult to standardize between test laboratories. In addition, when automatic measurement is assumed, there is a problem in quantification and reproducibility because it is not easy to distinguish between a nuclear region and a cytoplasmic region depending on the current staining method. Toxicity of a test substance is produced by fusing a gene encoding a single or a plurality of nuclear-related proteins and a gene encoding a fluorescent protein into a cultured cell to produce a mother cell expressed in the cell. Alternatively, by performing treatment for mutagenicity tests, performing limited excitation such that the micronucleus region emits fluorescence in a limited manner, and measuring the amount of fluorescence according to the nuclear-related protein component quantitatively. Provided is a method for detecting intracellular micronuclei, which enables detection in living cells without fixing the cells of interest. [Selection figure] None |
