abstract |
The present invention provides a recombinant cell obtained by a method of inducing at least one double-strand break in a site-specific manner in chromosomal DNA. An isolated DNA encoding an I-SceI enzyme is provided. The DNA sequence can be introduced into cloning and expression vectors, transformed cells and transgenic animals. The vector is useful for gene mapping and site-specific introduction of genes. Also provided is a recombinant cell that induces at least one site-specific break in chromosomal DNA and transforms the hair by a method of inserting a DNA encoding a polypeptide (a) having an I-SceI restriction site And (b) transforming said cell with at least one plasmid comprising DNA encoding I-SceI; and (c) at least one double-stranded DNA.
