abstract | PROBLEM TO BE SOLVED: To provide a matched fluorescent dye set having a molecular structure and charge adapted so that each dye can be covalently bound to a protein and the relative mobility in electrophoresis of the protein labeled with each dye is the same. The matched set includes two or more different fluorescent dyes of the formula. (Wherein n is 1, 2 or 3, Z 1 and Z 2 independently represent a group of carbon atoms necessary to complete the phenyl or naphthyl ring system, one of R 1 and R 2 groups is the target A linking group, the remaining R 1 or R 2 group is selected from — (CH 2 ) 4 —W or — (CH 2 ) r —H, wherein either R 1 or R 2 is — (CH 2 ). Except when it is r- H (where R 3 is W), the R 3 group is hydrogen and W is selected from sulfonic acid and sulfonate. Also provided is a method of saturating a protein with a fluorescent dye so that all of the amino acid (preferably cysteine) residues are labeled and a single population of labeled protein molecules is obtained. |
