abstract | Disclosed is a cervical-vaginal tissue equivalent consisting of vaginal epithelial cells and immune cells cultured at an air-liquid interface. Tissue equivalents can be infected with sexually transmitted pathogens such as viruses (eg, HIV), bacteria, parasites or fungi. Tissue equivalents can also perform allergic or stimulatory reactions. The tissue equivalent has a nucleated basal layer cell and a nucleated basal layer cell, and further has a cell layer outside the basal layer that gradually increases the content of glycogen and decreases the content of the nucleus. It is characterized by that. Tissue equivalent immune cells are initially located in the basal and suprabasal layers. Also disclosed is a method for making a tissue equivalent. The method includes providing vaginal epithelial cells and immune cells, seeding the cells on a porous support, and co-culturing at an air-liquid interface under conditions appropriate for differentiation. One such method disclosed is the creation of tissue equivalents in serum-free media. Certain cells in which tissue equivalents are formed and also certain preferred components of the medium in which tissue equivalents are produced are provided. Also disclosed are cervical-vaginal tissue equivalents made by the methods disclosed herein. |
