abstract | Disclosed are methods for modifying therapeutic compounds such as drugs that are substrates of active transporters expressed in epithelial cells lining the human colon lumen. Transporters expressed in the human colon include sodium-dependent multivitamin transporter (SMVT) and monocarboxylic acid transporters 1 and 4 (MCT 1 and 4). The modified compounds may themselves be pharmacologically active or may be metabolized to form pharmacologically active compounds as a result of cleavage of the chemical modification by uptake from the colon (e.g., Prodrug). The modified compounds disclosed herein are suitable for use in sustained release oral dosage forms, particularly those that release the drug over a period of time greater than about 2-4 hours after administration. |
