abstract | A method of detecting a change in the conformation of a nucleic acid sample by treating with an oxidizing agent (for example, KMnO 4 ) and determining the difference in the production or production rate of the reaction product MnO 2 or the consumption or consumption rate of KMnO 4 . The presence of a difference means a change in the conformation of the test sample, especially when the DNA is exposed to, for example, environmental salt concentrations and temperature or current. In the method of the present invention, for example, a change in DNA conformation when DNA is exposed to an intercalating agent is examined. Also described is the use of the method of the invention to examine DNA conformational changes in compounds. Such conformational changes are determined by a colorimetric inorganic assay that measures absorbance with a spectrophotometer. The use of OsO 4 or chromic acid or ozone gas, or peroxides or perbenzoic acid or other oxidants other than manganate, including current, is described herein. |
