The present invention provides amidases, polynucleotides encoding the amidases, and methods of making and using the polynucleotides and polypeptides. In one aspect, the invention provides enzymes having secondary amidase activity, such as enzymes having activity in amide hydrolysis, including enzymes having peptidase, protease and / or hydantoinase activity. In another aspect, the enzymes of the present invention are used to enhance food flavor (eg, enzyme-ripened cheese), promote sterilization and fungicides, modify and deprotect fine chemical intermediates, and synthesize peptide bonds. Chiral separation can be performed to hydrolyze cephalosporin C. The enzymes of the present invention can be used to produce 7-aminocephalosporanic acid (7-ACA) and semisynthetic cephalosporin antibiotics, including cephalosin, cephaloridine and cefuroxime. The enzyme of the present invention can be used as a bactericidal agent, for example, as a cell wall hydrolyzing agent. The present invention also provides a fluorescent amidase substrate comprising 7- (ε-D-2-aminoadipoylamido) -4-methylcoumarin. |
