abstract | The inventors of the present invention have discovered that asparagine synthase, 5-aminolevulinate synthase, histidinol-phosphatase, 3-isopropylmalate dehydratase and threonine synthase are each essential for fungal virulence. Specifically, inhibiting asparagine synthase, 5-aminolevulinate synthase, histidinol-phosphatase, 3-isopropylmalate dehydratase or threonine synthase gene expression in fungi does not result in any sign of successful infection or lesion. Thus, asparagine synthase, 5-aminolevulinate synthase, histidinol-phosphatase, 3-isopropylmalate dehydratase and threonine synthase can be used as targets for identifying antibiotics, preferably antifungal agents. Accordingly, the present invention provides methods for identifying compounds that inhibit asparagine synthase, 5-aminolevulinate synthase, histidinol-phosphatase, 3-isopropylmalate dehydratase or threonine synthase expression or activity. The method of the present invention is useful for identifying antibiotics, preferably antifungal agents. |
