abstract | Disclosed are large-scale virus production methods, particularly large-scale adenovirus production methods. The methods described herein are preferably applied to suspension cultures of mammalian host cells in large scale bioreactors where gas sparging is essential to provide sufficient aeration during culture. This method involves the use of high concentrations of compounds that protect host cells from the effects of gas sparging and agitation, and virus seed stocks made without cytolytic reagents. The present invention also provides an expandable method of making said viral seed stock and the resulting unclarified viral seed stock, which is concentrated to reduce storage capacity for infection of large-scale cultures and cells It also relates to those that do not contain dissolved components, such as the detergent Triton X-100 or polysorbate 80. |
