abstract | The object of the present invention is to assess the quality of ruminant feed and consequently prevent the transmission of TSE through the detection of animal proteins in these foodstuffs. This object is embodied in the form of a method for detecting protein of animal origin in a complex mixture, which comprises the following steps: (i) a high concentration of protein material from a sample of the initial complex mixture (Ii) preparation of matrix-analyte to maintain low levels of contaminants and proper matrix-analyte molar ratio; iii) Analysis of the material obtained in the previous step by MALDI-TOF mass spectrometry; (iV) optionally by isolating the fraction of the sample or its components by RP-HPLC and automatic sequencing of its N-terminal region Identification of said component, and sequencing of its peptide fragment by liquid chromatography coupled with mass spectrometry (LC / MS / MS). The present invention contemplates the use of this method in the detection of animal-derived proteins in ruminant diets, allowing the transmission of contagious spongiform brain injury, more specifically bovine spongiform brain injury (BSE).
