abstract | The present invention provides methods and compositions for imaging gene expression in cells. The composition of the present invention interacts with a target messenger RNA precursor molecule (target pre-mRNA) expressed in a cell, resulting in the generation of a novel chimeric RNA molecule (chimeric RNA) capable of encoding a reporter molecule. Contains a pre-trans-splicing molecule (PTM) designed to mediate the trans-splicing reaction. The PTMs of the present invention are designed to interact with the target pre-mRNA, thereby providing a method for detecting target pre-mRNA expression. The methods and compositions of the invention can be used to monitor the expression of specific genes in cells. Where specific gene expression is associated with a disease, the present invention provides diagnostic methods and compositions. Examples of such diseases include infectious diseases, proliferative diseases such as cancer, genetic diseases, neurological diseases and metabolic disorders. Furthermore, the present invention can be used in screening assays to identify compounds that can modulate gene expression, or assays designed to identify protein / protein interactions. The present invention is demonstrated by an example in which papillomavirus gene expression in a cell is detected using a bioluminescence assay system. |
