The present invention provides methods and compositions for gene expression analysis using nicking agents. The present invention provides a method for determining the presence or absence of a target cDNA molecule in a cDNA population or a target mRNA molecule in a biological sample, which method comprises the following steps: (A ) (I) a cDNA molecule of a cDNA population or an RNA molecule of a biological sample, (ii) a template nucleic acid molecule, (iii) a nicking agent that recognizes a recognition sequence, (iv) a DNA polymerase, and (v) one or more Forming a mixture comprising: deoxynucleoside triphosphates; (B) maintaining the mixture in conditions under which the single-stranded nucleic acid molecule is amplified; and (C) the presence or absence of the single-stranded nucleic acid molecule. Detecting the presence and determining the presence or absence of a target cDNA molecule in a cDNA population, or the presence or absence of a target mRNA in a biological sample The step of determining the presence. |
