titleOfInvention | Chemical peptide ligation with three or more components |
abstract | The present invention provides a method for constructing oligopeptide intermediates in a natural chemical ligation reaction that eliminates self-ligation of bifunctional intermediates. An important aspect of the present invention is a bifunctional intermediate having an N-terminal heterocyclic protecting group that effectively prevents self-ligation in the chemical assembly process. The present invention is useful in convergent synthesis of polypeptides and proteins, particularly when three or more peptide fragments are used to construct a polypeptide or protein product, and the natural chemical ligation efficiency is improved.
