Disclosed are thiol-linked polymeric prodrugs, methods of making and using the same. When a sulfhydryl bond is used in combination with a benzyl elimination system, a prodrug is formed that regenerates the parent molecule using plasma enzymes in vivo. A preferred prodrug of the present invention is Formula (I) when S-MP is 6-mercaptopurine. |
