abstract | 【Task】 The present invention provides an improved method of preparing fetuin, which uses a chelating agent to remove inorganic ions such as zinc, calcium, barium and the like from fetuin, "Naked" fetuin is added with zinc acetate to give a product consisting mainly of fetuin-zinc. This improved method of preparing fetuin-zinc or over-added fetuin increases the effectiveness of inducing apoptosis in cancer cells 3-4 fold. From this specially prepared zinc-added fetuin, certain peptide fragments were obtained. This polypeptide strongly induced apoptosis in LNCaP (prostate cancer) cells and HT-29 (colon cancer) cells without affecting CCD18co (normal colon) cells. This in vitro tissue culture study demonstrated that the polypeptide was about 10 times more potent than its parent molecule (full length fetuin plus zinc) in inducing apoptosis. |
