titleOfInvention | Production and use of reprogrammed human somatic cell nuclei and autologous and syngeneic human stem cells |
abstract | Activated human embryos produced by therapeutic cloning can generate human totipotent and pluripotent stem cells from autologous cells to obtain therapeutic grafts. The present invention provides a method for producing activated human embryos that can be used to develop totipotent and pluripotent stem cells that can yield autologous cells or tissues suitable for transplantation. In one embodiment, the present invention provides a method of producing an activated human embryo by parthenogenesis, and in another embodiment, the present invention provides that the genetic material of differentiated human donor cells is a therapeutic graft. Production of activated human embryos by somatic cell nuclear transfer by reprogramming to form a diploid human pronucleus capable of directing the generation of said stem cells derived from autologous, syngeneic cells Provide a method. Said ability to create autologous human embryos represents an important step in generating immunocompatible stem cells that can be used to overcome the aforementioned problems of immune rejection in regenerative therapy. The activated human embryo produced by the present invention also provides a model system for identifying and analyzing the molecular mechanism of epigenetic imprinting and the genetic regulation of embryogenesis and development. |
