The present invention relates generally to variants of target biological molecules ("TBMs"), as well as methods of making them and methods of using them to identify ligands for TBMs. More particularly, the present invention relates to individual variants TBM and sets of variants TBM, each of which is a modified version of a protein of interest in which a thiol is introduced at or near the site of interest.
