abstract | An object of the present invention is to provide a screening method for inhibitors of γ-glutamylcysteine synthetase (γGCS) specific to parasites and the like. A human type γGCS in which the amino acid residue corresponding to the 132nd position of E. coli γGCS is arginine and a non-human γGCS in which the amino acid residue corresponding to the 132nd position is other than arginine are prepared. The drug candidate compound prepared in advance is brought into contact with each of the human-type and non-human γGCS to evaluate the binding strength, and the drug binds more strongly to the non-human γGCS than to the human-type γGCS Selecting a candidate compound. The design of the drug candidate compound used in the screening method of the present invention is a co-crystal of Escherichia coli γGCS and a substrate transition state analog, and is based on the three-dimensional structure data of γGCS obtained by analyzing a crystal whose space group is P2 1.
