abstract | [Problem] To provide a method for preparing high-purity DNA having the same molecular weight and retaining double strands, and DNA as a functional material. A high-purity polymer DNA is enzymatically digested with an enzyme that cleaves a DNA strand, and after cleaving to a DNA having a desired molecular weight, is further added with an enzyme that specifically cleaves single-stranded DNA. As a functional material characterized in that the single-stranded DNA is cleaved by enzyme treatment and the cleaved single-stranded DNA is removed, so that the target molecular weight is aligned and the percentage of double-stranded DNA is increased. This is a method for preparing DNA. Also, a DNA solution and a lipid solution as a functional material obtained by the preparation method are mixed and stirred to obtain a DNA-lipid complex, which is melted in a pressure-bonded or organic solvent by heat and extended so as to spread.
