PROBLEM TO BE SOLVED: To provide a novel process that is useful and applicable for nucleic acid amplification, nucleic acid sequencing, and generation of unique nucleic acids having important characteristics (eg, reduced thermodynamic stability). A process for linearly amplifying a specific nucleic acid sequence comprising the following steps: the specific nucleic acid sequence, initial primer or nucleic acid construct comprising two segments Providing a construct; and a substrate, buffer, and template-dependent polymerizing enzyme; and in the presence of the substrate, buffer, and template-dependent polymerizing enzyme under equilibrium or limited cycling conditions, Incubating a specific nucleic acid sequence and the novel primer or nucleic acid construct; thereby linearly amplifying the specific nucleic acid sequence.
