abstract |
An object of the present invention is to provide a production method capable of efficiently giving a product having high optical purity in the production of (R) -2-chloromandelic acid amide. Another object of the present invention is to provide a novel method for producing an α-ketoamide reductase that uses NADPH as a coenzyme to reduce 2-chlorobenzoylformamide to produce (R) -2-chloromandelic acid amide with high optical purity. To do. Among the enzymes having 2-chlorobenzoylformamide reducing activity present in Saccharomyces cerevisiae, an enzyme with high stereoselectivity was purified and its enzymatic scientific properties were clarified. By analyzing a part of the internal amino acid sequence of the purified enzyme, we found that this enzyme is encoded by the predicted open reading frame (ORF) YDL124w reported by genome analysis. As a result of cloning and expressing YDL124w in E. coli, it was clarified that this ORF encodes α-ketoamide reductase. It was found that (R) -2-chloromandelic acid amide can be produced from 2-chlorobenzoylformamide by the obtained transformant. [Selection figure] None |
