abstract |
The present invention is a method for characterizing the metabolic stability of a drug candidate, wherein the nuclear polarization of an NMR active nuclide present in the drug candidate is increased (hereinafter referred to as “hyperpolarization”), and the drug candidate is defined as a hepatocyte. Or it relates to a method comprising contacting with an organelle and performing NMR analysis. [Effect] This method is a rapid and simple in vitro assay and is particularly suitable for screening a large number of drug candidates. The methods of the present invention do not require isolation of drug candidates and / or their metabolites from the assay material. Since the hyperpolarized drug candidate is used in the method of the present invention, the sensitivity is superior to that of a normal NMR assay, and therefore, a very small amount of metabolites that cannot be detected by conventional NMR can be detected.
