abstract PROBLEM TO BE SOLVED: To provide an immortalized mast cell line possessing the functions and characteristics inherent in mast cells, its establishment and its establishment method, and a screening method for useful substances using such immortalized mast cell line. SOLUTION: A bone marrow cell of a transgenic mouse into which a large T antigen gene of SV40 temperature-sensitive mutant tsA58 has been introduced is hemolyzed, and then the resulting cell is cultured in the presence of IL-3 to obtain mast cells. Differentiation / induction, repeated subculture, FcεRI and c-kit molecules are expressed on the cell surface, granules are abundantly stored in the cytoplasm, and activated by co-crosslinking of FcεRI and degranulation ability and Ca 2+ A cell line having the ability to induce increase in concentration is established.
