abstract | The living biological material is cryopreserved by immersing the material in a tank of coolant and circulating the coolant past the material at a substantially constant predetermined rate to freeze the material. Prior to freezing, the material is directly immersed in a cooling liquid without treatment or chemically treated. The method of the present invention freezes the biomaterial quickly enough to avoid the formation of ice crystals within the cellular structure, maintains its anatomical structure for the sample, and preserves the biochemical activity after freezing. The temperature of the coolant is preferably between −20 ° C. and −30 ° C., a temperature that is sufficiently warm to minimize the occurrence of stress breakdown and the formation of artifacts in the cell membrane due to thermal fluctuations. Cells frozen using the method of the invention had significantly less cell-to-cell damage than cells cryopreserved by other cryopreservation techniques used in pathological and histological procedures. Because the present invention allows the biomaterial to be frozen such that the material vitrifies, the intracellular biochemical activity does not disappear after freezing, and various embodiments of the present invention are useful in the field of research and patient care. May be used in devices for preparing biomaterials for new approaches to cryohistology and immunohistochemistry. |
