abstract |
The intracellular tumorigenic chemistry of the molecule is measured by using a fluorescently labeled substrate that changes its electrophoretic mobility upon a chemical reaction, such as a chemical reaction caused by an enzyme or kinase. Specificity is achieved by using a labeled substrate molecule that reacts only to specific tumor enzymes. At the point of interest, the presence of such a substrate molecule in the cell is measured. To ensure accuracy, measurements must be made in a timely manner to minimize chemical reactions that occur subsequent to the point of interest. A controllable rapid laser lysis method is used to obtain the contents of the cells into which the reporter substrate molecule has been introduced. The contents of the cells are then subjected to capillary electrophoresis, and the activity of the tumorigenic enzyme is separated by electrophoresis and the amount of unaltered substrate molecule and the altered substrate molecule identified by the presence of a fluorescent label.
