abstract | The method for measuring the extent of the forward nucleic acid polymerase reaction to produce pyrophosphate can be performed in the presence of all components necessary for the progress of nucleic acid synthesis, which can be either dATP or dATP analog. A nucleic acid polymerase reaction substrate. The method includes pyrophosphate analysis comprising the steps of converting pyrophosphate to ATP and detecting light produced by the bioluminescent reaction of luciferase and ATP, one or both of the following being applicable: a) reacting luciferase with ATP and substrate, the spectral overlap is reduced compared to the spectral overlap between the output of the reaction of wild-type Photinus pyralis luciferase and ATP and dATP, respectively; b) The luciferase reacts with the substrate, resulting in reduced bioluminescence compared to the luminescence produced by the reaction of wild-type Photinus pyralis luciferase with dATP. |
