abstract | Kind Code: A1 Abstract: Alfin 1 cDNA encodes a putative transcription factor associated with salt tolerance in Medicago sativa L. The recombinant protein specifically binds to DNA including a promoter fragment of the salt-inducible gene MsPRP2. A callus that overexpresses Alfin 1 is more resistant to growth inhibition by 171 mM NaCl than a vector transformation control, whereas a callus that expresses Alfin 1 antisense is more sensitive to salt inhibition. there were. Transgenic plants that overexpress Alfin 1 in the sense orientation grew well. In contrast, antisense transgenic plants grow poorly in soil, indicating that Alfin-1 expression is essential for normal plant development. Transgenic calli and plant roots overexpressing Alfin 1 showed elevated levels of endogenous MsPRP2 mRNA accumulation. However, MsPRP2 mRNA accumulation was also regulated in a tissue-specific manner as shown in leaves of transgenics overexpressing Alfin 1. These results also suggest that alphin 1 acts as a transcriptional regulator in MsPRP2 expression in purple palmetto and plants.
