Peptides that bind cellular (surface or intracellular) receptors, such as nuclear receptors, can be identified by screening a combinatorial peptide library expressed in cellular form, along with a signal generation system to report binding. The cells co-express one member peptide and the receptor. The "two-hybrid" assay is of particular interest. The screen is performed in the presence of a ligand, especially an exogenous ligand. If screening is performed for multiple different receptor conformations, this library screening will also help identify conformation specific peptides for the receptor, and then interact with the receptor in the presence of each panel peptide Used in panels to "fingerprint" compounds in question as their ability to do so. These fingerprinting can compare those of the reference compounds to known bioactives mediated by the receptor. |
