abstract The present invention includes a water-in-oil emulsion, wherein the water-in-oil emulsion has the general formula A-COO-B-OOC-A (wherein component B is a divalent residue of a water-soluble polyalkylene glycol). And component A provides an adjuvant for use in the preparation of a vaccine, characterized in that it comprises a polymer emulsifier which is a block copolymer having a residue of an oil-soluble complex monocarboxylic acid. Such polymeric emulsifiers and their preparation are disclosed in British Patent No. 2002400 and WO 9607688. Preferably, in a block copolymer of the formula A-COO-B-OOC-A, component B is derived from polyethylene glycol and component A is derived from polyhydroxystearic acid, preferably poly (12-hydroxy-stearic acid). Be guided. The most preferred emulsifier used in the adjuvant of the present invention is Arlacel P135, PEG30 dipolyhydroxystearate. Another similar emulsifier for use with the present invention is ATLOX® 4912. Arlacel P135 and ATLOX® 4912 are both block copolymers (ABA) of polyethylene glycol and polyhydroxystearic acid having a molecular weight of about 5000, commercially available from ICI. It has been found that these emulsifiers are used to obtain adjuvants based on stable, low viscosity w / o emulsions that may be based on metabolic oils. The adjuvant has excellent adjuvant activity and is well tolerated.
