abstract |
The present invention provides a system for producing differentiated cells from the stem cell population used, wherever a relatively uniform cell population is desired. Such cells contain an effector gene placed under the control of a transcription control element (such as the TERT promoter) that causes the gene to be expressed in relatively undifferentiated cells of the population. Expression of the effector gene either depletes the undifferentiated cells or expresses a marker that can be used later to remove the undifferentiated cells. Suitable effector sequences encode enzymes (such as thymidine kinase) that convert toxins, apoptosis-inducing proteins, cell surface antigens, or prodrugs into substances that have lethal effects on cells. Differentiated cell populations generated in accordance with the present disclosure are suitable for use in tissue regeneration and non-therapeutic applications (such as drug screening). |
