abstract Under the condition that both ends of at least one end of the first double-stranded nucleotide sequence (ds) are covalently linked to both ends of at least one end of the second ds nucleotide sequence by topoisomerase, A method is provided for making a double-stranded recombinant nucleic acid molecule in which both strands are covalently linked by contacting an additional ds nucleotide sequence with topoisomerase. Similarly, two or more ds nucleotide sequences may be of type IA, provided that one strand at one or both ends of the first ds nucleotide sequence is covalently linked by topoisomerase, but not both. Also provided is a method of making a ds recombinant nucleic acid molecule having one strand covalently linked thereto by contacting with topoisomerase. Compositions for performing such methods, and compositions resulting from such methods, are also provided, as are kits containing components useful for practicing the methods conveniently.
