abstract The present invention belongs to the field of thrombolysis and the field of tissue plasminogen activator (tPA) derivative production in prokaryotic cells. The present invention provides a method for producing a recombinant DNA-derived tPA, a mutant thereof or a (Kringle 2 serine) K2S molecule or a mutant thereof in prokaryotic cells, wherein the tPA or K2S or the mutant is active and correctly folded. Said prokaryotic cell is secreted extracellularly as a transcribed protein, said prokaryotic cell containing and expressing a vector comprising said tPA or K2S or a DNA encoding a mutant operably linked to a DNA encoding a signal peptide OmpA. About. The invention further relates to certain K2S derivatives obtainable by said method. The invention further relates to said DNA molecule and the use of said DNA molecule in said method.
