abstract | A triple-stranded complex includes a single-stranded probe bound to a double-stranded nucleic acid target, the single-stranded probe consisting of a heteropolymer nucleic acid or a heteropolymer nucleic acid analog. All base triplets of the complex are ATA, TAT, UAT, TAU, AUA, UAU, GCG. And C-G-C. Cation-assisted assays involve detecting the presence of such a triple-stranded complex to determine the degree of complementarity between the probe and the target sequence. The assay preferably detects a change in the fluorescence intensity of the label as a function of the binding affinity between the probe and the target. The label may be covalently attached to the probe or target, or it may be an insertion fluorophore in the reaction medium. |
